Abstract:【Objective】 To evaluate the diagnostic value of high-frequency ultrasound-guided core needle biopsy combined with tumor marker testing for distinguishing benign from malignant breast masses. 【Methods】 A total of 98 patients with breast masses admitted to our hospital between January 2021 and June 2025 were enrolled. Among them, 51 patients diagnosed with breast cancer by postoperative pathological examination were assigned to the observation group, and 47 patients with benign breast lesions were assigned to the control group. All patients underwent high-frequency ultrasound-guided core needle biopsy and measurement of carbohydrate antigen 125 (CA125), carbohydrate antigen 15-3 (CA15-3), and alpha-fetoprotein (AFP) levels. The diagnostic performance of the combination of high-frequency ultrasound-guided core needle biopsy with CA125, CA15-3, and AFP was assessed using receiver operating characteristic (ROC) curves. 【Results】 High-frequency ultrasound-guided core needle biopsy revealed 47 cases of breast cancer and 44 cases of benign breast lesions. The levels of CA125, CA15-3, and AFP in the observation group were significantly lower than those in the control group (P<0.05). ROC curve analysis showed that the sensitivity and specificity of high-frequency ultrasound-guided core needle biopsy combined with detection of CA125, CA15-3, and AFP for diagnosing benign versus malignant breast masses were 94.1% and 97.9%, respectively, with an area under the ROC curve (AUC) of 0.982. 【Conclusion】 The combination of high-frequency ultrasound-guided core needle biopsy with CA125, CA15-3, and AFP testing significantly improves the sensitivity and specificity in diagnosing the nature of breast masses and holds substantial clinical value.
