Abstract:【Objective】 To detect the expression of lncRNA BC002811 in gastric cancer tissues and cell lines, and to study its function regulating the migration and invasion in gastric cancer cells. 【Methods】 Real-time PCR was used to detect the expression of BC002811 in gastric cancer tissues and cell lines. Gastric cancer cell stable strains with overexpression and down-regulation of lncRNA BC002811were established. Subsequently, the migration and invasion capability of different strains were analyzed with Scratch (wound-healing) test and transwell assay, respectively. Finally, real-time quantitative PCR and Western blot were used to investigate the molecular mechanism of BC002811 to promote tumor progression.【Results】 The relative expression of lncRNA BC002811 was significantly up-regulated in gastric cancer tissues and cell lines compared to corresponding adjacent normal tissues and normal gastric cell line, respectively(P<0.05). When lncRNA BC002811 was over-expressed, the migration and invasion ability of gastric cancer cells were enhanced and the expression of matrix metalloproteinase-2 (MMP2), MMP9 and Vimentin (one of the mesenchymal cell markers) increased. While the expression of E-cadherin (one of the epithelial cell markers) decreased(P<0.05). When the expression of lncRNA BC002811 was inhibited, the opposite results occurred. 【Conclusion】 lncRNA BC002811 promoted the migration and invasion in gastric cancer cells and contributed to the tumor deterioration.
