Abstract:【Objective】To observe the effect of siRNA silencing VDAC1 gene on the proliferation and cell cycle of breast cancer MCF-7 cells.【Methods】siRNA was designed and synthesized for human VDAC1 gene and MCF-7 cells were transfected through VDAC1-siRNA. Real time fluorescent quantitative PCR and Western blot were used to detect the expression level of VDAC1 in MCF-7 cells;MTT assay was used to detect the proliferation of MCF-7 cells, the cell cycle distribution of MCF-7 cells was detected by flow cytometry, and the expression of cyclin D1 was detected by Western blotting【Results】VDAC1-siRNA transfected from MCF-7 cells can effectively inhibit VDAC1 expression, inhibit the proliferation level, promote MCF-7 cells to occur G1 phase arrest and down regulate the expression of Cyclin D1 protein.【Conclusion】VDAC1 silencing can effectively inhibit the proliferation of human breast cancer MCF-7 cells, and promote cell arrest in G1 phase, which can be used as a new target for the treatment of breast cancer.
