Abstract:【Objective】 To investigate the optimal dosage of STZ (streptozotocin) in inducing diabetic itch in C57BL/6 mice. 【Methods】Twenty-four male aged 8 weeks old C57BL/6 wild type mice (the weight 20~28 g) were randomly divided into 4 groups. A. Control group (mice were given the same dose solvent by intraperitoneal injection); B. High dose group (mice were intraperitoneally injected with STZ 160mg/kg); C. Low dose group (mice were intraperitoneally injected with STZ 40mg/kg 5 times continuously and the interval time was 24 hours); D. Mild moderate dose group (mice were intraperitoneally injected with STZ 85mg/kg + 65mg/kg and the interval time was 24 hours). Mice in the Group A were fed with normal food and B、C、D groups mice were fed with high-fat food from 4 weeks before the injection to the end of experiment. At the time of 1D before the injection and 1,2,3,4 weeks after injection, the fasting blood glucose, body weight and scrathing bouts in 30 minutes were detected. 【Results】Compared with the group A, the blood glucose level of other groups increased to the diabetic standard(>200mg/dl)at different time point (P<0.05). The weight of the group B was lower than that of the group A, group C and group D (P<0.05), and the number of scratching in the group D was increased more than that of A, B and C at second, third weeks after STZ injection (P<0.05).【Conclusions】It is a suitable way to establish the diabetic itch model of mice induced by intraperitoneal injection 85mg/kg+65mg/kg STZ combining with feeding high-fat food.
