Abstract:【Objective】To investigate the effect of low molecular weight heparin (LMWH) on the proliferation and the SDF-1expression of primary cultured gastric carcinoma cells. 【Methods】Poorly differentiated adenocarcinoma of the stomach tissues and normal gastric mucosa tissues were made into primary cells and then divided into four groups:group A had gastric cancer cells, group B had gastric cancer cells + LMWH, group C had gastric mucosal cells, and group D had gastric mucosal cells + LMWH. The cell proliferation was detected by MTT assay, the cell cycle distribution and apoptosis was detected by V-FITC/PI assay, and the expression of SDF-1 mRNA was examined by semi-quantitative RT-PCR.【 Results】 The proliferation of gastric carcinoma cells in group B was much lower than in group A. At the same time, cell apoptosis was higher in group B than in group A. In the G0 and Gl phases,there was a higher ratio of cells but a lower ratio was observed in the S period. There was no significant difference in cell ratios in the G2 and M phases between these two groups.For groups C and D, there was no significant difference for cell proliferation, cell cycle, or apoptosis. The expression of SDF-1 mRNA was lower in group B than in group A. There was no significant difference in the expression of SDF-1 mRNA in groups C and D. 【Conclusion】LMWH can inhibit the proliferation of primary gastric carcinoma cells and also increase the cell apoptosis, changing the structure of the cell cycle and reducing the expression of SDF-1 mRNA in primary cells of gastric cancer.
