Abstract:【Objective】To explore the protection mechanism of peroxidase 6 (PRDX6) against liver ischemia reperfusion injury. 【Methods】A liver ischemia reperfusion injury model was established. And normal control group, sham group, ischemia reperfusion injury group, and quercetin group were set. Liver tissue was collected from all groups and total proteins were analyzed, separated by twodimensional gel electrophoresis and identified by matrixassisted laser desorption ionization time of flight mass spectrometry, PRDX6 was verificated by immunohistochemical staining. 【Results】Twodimensional gel electrophoresis showed that there were about 825±29 protein spots in the ischemia reperfusion injury group, and 831±27 protein spots in quercetin group. There was a 44 protein spot difference between the two groups, 25 were being upregulated, and 19 being downregulated. Experimental results suggest quercetin can induce the expression of PRDX6, and the highest expression was at 12 h after reperfusion. 【Conclusion】PRDX6 may be related with protection of quercetin against liver ischemia reperfusion injury, which is beneficial for the understanding of the mechanism of protection of quercetin against liver ischemia reperfusion injury.
