Abstract:[Obj ective]To explore the expression of immunologic receptor nucleotide-binding oligomerization domain-containing protein 1(NOD1)in adipocytes of obese patients with diabetes mellitus(DM)and its relation with insulin re-sistance(IR).[Methods]Abdominal subcutaneous adipocytes were drawn from 10 obese patients with DM(DM group) and 1 0 obese patients without DM(NDM group).Real-time PCR was used to detect the expression of NOD1 in adipocytes of two groups.NOD1 receptor agonist iE-DAP was administered.Enzyme linked immunosorbent assay(ELISA)was used to detect the levels of inflammatory cytokines such as interleukin-6(IL-6),interleukin-8(IL-8)and monocyte chemotactic factor-1(MCP-1)in two groups.The baseline state and iE-DAP-stimulated glucose uptake were observed by 2-deoxy-D-3 H glucose uptake assay.[Results]NOD1 mRNA expression in adipocytes of DM group was markedly higher than that of NDM group(P <0.05).The levels pf IL-6,IL-8 and MCP-1 in DM group with iE-DAP stimulation were markedly higher than those in NDM group(P<0.05).Adipocyte glucose transport rate in DM group with iE-DAP stim-ulation was significantly lower than that in NDM group(P<0.05).[Conclusion]NOD1-mediated inflammatory response may involve in the occurrence of IR in obese DM.
