摘要【目的】探讨转化生长因子β1(TGFβ1)对人胚肺成纤维细胞中微小核糖核酸-29(miR-29)表达的影响。【方法】以未处理组为对照,U6基因做内参,用荧光定量PCR(real-time PCR)法检测 TGFβ1处理的人胚肺成纤维细胞(IMR90)中miR-29a/b/c的表达。【结果】用浓度为1,2,4 ng/mL的TGFβ1处理IMR90细胞时,与对照组比较,其miR-29a/b/c相对表达依次降低( P <0.05);用浓度为2 ng/mL的TGFβ1分别处理IMR90细胞24 h ,48 h ,96 h ,与未处理组比较,其miR-29a/b/c相对表达逐渐下调( P <0.05)。【结论】在IMR90细胞中,TGFβ1可抑制miR-29的表达,预示 TGFβ1-miR-29通路在肺间质纤维化形成过程中起重要作用。
Abstract:[Objective]To explore the impact of transforming growth factor beta-1(TGFβ1 ) on the expres-sion of micro-RNA-29(miR-29) in human embryonic lung fibroblasts(IMR90) .[Methods]The expression of miR-29a/b/c in IMR90 cell line treated with TGFβ1 was detected by fluorescence quantitative PCR (real-time PCR) assay normalized to the blank cell and U6 .[Results]When IMR90 cell line was treated with 1ng/mL , 2ng/mL and 4ng/mL TGFβ1 ,the expression of miR-29a/b/c was in turn decreased in comparison with control group ( P <0 .05) .When IMR90 cell line was treated with 2ng/mL TGFβ1 for 24 ,48 and 96h ,the relative expression of miR-29a/b/c was gradually decreased in comparison with the untreated group ( P < 0 .05) .[Conclusion] In IMR90 cell line ,TGFβ1 can inhibit the expression of miR-29 which indicates that TGFβ1-miR-29 pathway may play an important role in the formation of pulmonary interstitial fibrosis .
